Question : Problem: How to reconfigure Serial port?

I have a PC I use as a cash Register I disconnect the cash register Read out which is a display device for the customer to see the prices, It is connected through serial cable connected to an adapter to convert to US B connection since the only serial port came with the PC is Busy, After I disconnect that device I connected again but is not configured what should I do all the drivers are there but it seem it lost it's own port Please advice
